Q. "If Jesus' birth was, as we believe, by immaculate conception, why do we refer to Him as the seed of Jesse, of the line of David?"

Answer: Joseph seems to have been the last remaining heir to the throne of David. He accepted Mary as his wife, and then had the baby Jesus recorded in the Bethlehem census as his son. That gave Jesus the right to be the king from the line of David. See Matthew 1:18-25 and Luke 2:3-5.   This was apparently never questioned  by the Jewish authorities.
